The first structure of an Aldehyde
dehydrogenase reveals novel interations between NAD and the Rossmann fold
Nature Structural Biology, 1997, 4(4), 317-326
A. Introduction to aldehyde dehydrogenase
Aldehyde DHase (ALDH) is an important emzyme in
alcohol metabolism , ALDH make acetaldehyde
convert into acetic acid, and these step (alcohol DHase, and aldehyde
DHase) is " detoxication"steps in liver.
(II)And it exists as distinct enzymes with different tissue specificities, eyes , liver, kidney.....
Aldehyde DHase (ALDH) has three classes:
class¢¹: cytosolic ALDH
class¢º: mitochondrial ALDH
class¢»: tumor associated
(In this paper"Nature structural biology"
is Class ¢» in rat liver)
(III)Aldehyde DHase is homodimer, and have three domains:
1. NAD-binding domain
2. Catalytic domain
3. Bridging domain

(IV)Topology of ALDH
and its topology contains Rossmann
