The first structure of an Aldehyde dehydrogenase reveals novel interations between NAD and the Rossmann fold

A.  Introduction to aldehyde dehydrogenase

Aldehyde DHase (ALDH) is an important emzyme in  alcohol metabolism , ALDH make acetaldehyde
convert into acetic acid, and these step (alcohol DHase, and aldehyde DHase) is " detoxication"steps in liver.

(I)ALDH catalytic mechanism

(II)And it exists as distinct enzymes with different tissue specificities, eyes , liver, kidney.....

Aldehyde DHase (ALDH) has three classes:
    class¢¹: cytosolic ALDH
    class¢º: mitochondrial ALDH
    class¢»: tumor associated

(In this paper"Nature structural biology" is Class ¢» in rat liver)

(III)Aldehyde DHase is homodimer, and have three domains:
       1. NAD-binding domain
       2. Catalytic domain
       3. Bridging domain

(IV)Topology of ALDH

and its topology contains Rossmann fold(£]-£\-£])
