Oncology, Cell Biology. The p53 gene expression is as important response checkpoint in cells during stress environment. The high-risk HPVs E6 protein is thought to bind P53, promoting it degradation through the ubiquitin pathway and thus abrogates its function. Previously, we have found arsenite or cisplatin treatment could restore wild type (but not mutant) p53 expression, its downstream p21 and mdm2 gene expression, enhance apoptosis in several E6 containing mammalian cells. Our continue studies still focus on the effects of E6 and E7 oncogene and other environmental factors (such as virulent or virulent-associated factors) on P53 and its downstream genes regulation. Through this study, we may provide another possible strategy to promote some specific agents as another potential candidate for the therapeutic purpose in cancer or HPV-positive cancer cells.
Functional studies on unique specific gene products.? The complete genome sequence (1.6 Mb, open reading frames 1590) of Helicobacter pylori has been determined in 1997. Although itis relatively small and suitable for a structural genomics and proteomics studies, the relative not much studies had been reported.—Less than 40 proteins in proteomics or structural biology had been determined according to PDB record.? The functions of the pyrophosphatase and spermidine synthase in H. pylori were confirmed in our laboratory recently.? Their structures were solved in our collaborated laboratory already.? Since genomic DNA microarray results from other lab. indicated that many acid-inducible proteins were found in bacteria in pH 5.5 (in comparison to pH 7.2), we chose several acid-inducible target genes for further functional studies after constructing each gene into expression vector and purifying the protein from host E. coli.? Currently, we concentrated functional studies on 4 target proteins (acid-inducible: a unique virulent associated protein, NAD(P)H:FMN oxidoreductase, nucleotide metabolism related: UMP kinase and dUTPase).? For above virulent associated protein, we focus on biological functions, the gene expression on mRNA and protein level in H. pylori under various environmental conditions. We are examining the (increase or decrease the tested human gene expression) effects of this protein on human gastric adenocarcinoma cells.
