Upon addition of GdnHCl, both mutants exhibit a fast conformational unfolding when monitored by absorbance and fluorescence, as well as slow phase detected only by fluorescence which corresponds to the isomerizations of Pro93 and Pro114.

The slow fluoresence unfolding phase is independent of pH and [GdnHCl], and has an activation energy of 20kcal/mol, which strongly suggests that the slow phase correspons to the isomerization of Pro93 and Pro114.
