Specific transcription factors belong to a few families

  • Yeast transcription factor GAL4 contains a binuclear zinc cluster in its DNA-binding domain

  • - subfamily of zinc-containing motifs from yeast and fungi
    - activates transcription of genes required for break down of the sugar galactose and melibiose
    - large protein, 881 AA
    - DNA binding region 1-49
    - Dimerization region 50-94
    - Acidic activating regions 94-106, 148-196, 768-881

  • The zinc cluster regions of GAL4 bind at the two ends of the enhancer element

  • The linker region also contributes to DNA binding

  • - responsible for specificity
