Transportin-SR2 (MTR10) interacts

with hnRNPA1

Transportin-SR2 (MTR10) can interact with the phosphorylated RS dormain of several cellular SR proteins and participate in the trafficking of these SR protein into nucleus.

 

SR proteins are mainly involved in the splicing of precursor mRNA.

 

Using MTR10 as a bait in a yeast two-hybrid screen, it was found that hnRNPA1 interacts with MTR10.

 

HnRNPA1 includes a M9 domain and a RG box. M9 domain is an import/export signal and RG box has rich arginine and glycine.

 

We constructed pGAD-hnRNPA1-ΔM9 and pGAD-hnRNPA1-ΔRG in order to determine the interaction between MTR10 and hnRNPA1. Both RG box and M9 signal might be the domain interacting with MTR10.

We used a yeast two hybrid system and β-galactosidase assay to determine the result.
